What this is

A 16-amino-acid mitochondrial-derived peptide encoded within the mitochondrial 12S rRNA gene.

That sentence contains the single most interesting fact about MOTS-c, so let us slow down on it. Almost every peptide in this catalog is encoded in nuclear DNA — the genome in the cell's nucleus. MOTS-c is not. It is encoded inside the mitochondrial genome, a small separate loop of DNA that mitochondria carry from their own evolutionary history. A short open reading frame tucked inside the 12S ribosomal RNA gene codes for this 16-amino-acid peptide.

Peptides encoded this way are called mitochondrial-derived peptides, and there are only a handful of confirmed ones. We stock two: MOTS-c and Humanin, which comes out of the 16S rRNA gene instead. If you are working on mitochondrial signalling, those two are usually studied as a set.

Studied in metabolic and mitochondrial research as a model for AMPK-pathway signaling and cellular energy regulation.

AMPK is a cellular energy sensor — the enzyme complex that reads the ratio of spent to unspent energy currency in the cell and shifts metabolic behaviour accordingly. It is one of the most-studied nodes in metabolic biology, and MOTS-c is frequently used as a probe for it.

The broader research framing here is retrograde signalling: the idea that mitochondria do not only take instructions from the nucleus but send signals back. MOTS-c is one of the molecules used to investigate that direction of traffic, which is why the literature clusters around cellular energy regulation rather than any single organ or tissue.

Research on mitochondrial-derived peptides is comparatively young — MOTS-c was first described in 2015 — so the body of work is thinner than for, say, BPC-157. That is a reason to read closely, not a reason to avoid it. It also means most of what exists is in-vitro and animal work.

What does "mitochondrial-derived" actually mean?

The gene that codes for MOTS-c sits in the mitochondrial genome, not the nuclear genome. Mitochondria carry their own small circle of DNA, and this peptide is encoded inside the 12S rRNA gene on it.

Is MOTS-c related to Humanin?

They are siblings in the same small family. Humanin is also a mitochondrial-derived peptide, encoded in the 16S rRNA gene instead of the 12S. Researchers working on mitochondrial signalling usually look at both.

Which category does MOTS-c sit in?
